(1) Physical

The inclusion of β-CD with the drug is a physical process, no chemical reaction occurs, and the drug still retains its original properties and effects. Cyclodextrin has a hydrophobic cavity, which can selectively include various guest molecules, change the properties of the drug at the molecular level, and form a stable inclusion compound, thereby achieving the purpose of improving the water solubility and stability of the guest molecules. Hydroxypropyl cyclodextrin (HP-β-CD) has excellent water solubility, is non-toxic to the kidney, and has almost no stimulating effect on muscles and mucous membranes. Inclusion of poorly soluble drugs with HP-β-CD can significantly improve their stability, solubility and bioavailability.

 

(2) Good thermal stability

Cyclodextrin has good thermal stability. At about 100 ℃, when water evaporates from the crystal, it produces an endothermic effect; at about 270 to 280 ℃, when the crystal melts and thermal decomposition occurs, it produces an endothermic effect. In other temperature ranges, this crystalline material does not absorb heat. The secondary and tertiary structure of starch, protein and other macromolecular materials used for encapsulation will change when heated, and in most cases, this change is irreversible, so heating will cause the loss of polymer molecules

Capsule material capacity. The structure of cyclodextrin will not change after heating and cooling, maintaining the ability to form complexes. During the heating process, there is some loss of volatile guest, and the amount of loss depends on the nature and water content of the guest. For dry composites, the evaporation temperature of the guest often exceeds the temperature of the uncomposited guest by more than 10 ℃. If the guest contains water, some of the guest will exist in a free (uncomplexed) state, which is likely to cause loss when heated. Drugs and cyclodextrin inclusion complexes that are unstable in exposure to light, oxygen and heat are wrapped in the cyclodextrin cavity to avoid contact with external unstable factors and increase the stability of the drug.

 

(3) Other features

In addition to the above-mentioned characteristics, the preparation of the drug into a cyclodextrin inclusion compound also has the characteristics of liquid drug powderization, prevention of the volatilization of volatile components of the drug, and improvement of the bioavailability of the drug.
